The Swans turned in a five-star showing to crush the league leaders at the Matchroom Stadium.
In an unbelievable display, Roberto Martinez saw his side score five times without reply through a Tom Butler brace, Paul Anderson, Darren Pratley and Warren Feeney.
The result took the Swans up to fourth place and sent a clear message out to the rest of League One.
Leyton Orient:
Nelson, Purches (capt), Daniels, Thelwell, Mkandawire, Melligan, Chambers, Gray, Thornton, Boyd, Palmer. Subs: Corden, Morris (GK), Oji, Ibehre, Demetriou
Swansea City: de Vries, Rangel, Painter, Lawrence, Monk (capt), Butler, Bodde, Pratley, Anderson, Scotland, Feeney. Subs: Austin, O'Leary, Orlandi, Knight (GK), Allen.
Sixth-placed Swansea made just one change from the side that beat Swindon 2-1 at the Liberty Stadium in midweek.
With Darryl Duffy still suffering from a groin injury, Warren Feeney started alongside Jason Scotland up front after coming on to grab the winner against the Robins.
Midfielder Andy Robinson was given a few extra days to recover from his hamstring injury and didn't travel.
League leaders Orient made two changes from their 3-1 defeat at Gillingham with skipper Stephen Purches returning from a hamstring injury at right back to make it a real family affair.
The captain's brother-in-law is Swans striker Warren Feeney.
Charlie Daniels was also called up for his first league start in O's colours, with Paul Terry and Jason Demetriou making way.
The game started off at a frantic pace as Ferrie Bodde brought an easy save out of Stuart Nelson with a first minute free-kick.
As play switched immediately to the other end, Dorus de Vries made his save from Charlie Daniels after Orient had won a corner.
And after such a positive start, the breakthrough arrived after just three minutes as Swansea flew into the lead.
Bodde and Darren Pratley combined to find Tom Butler out wide and the midfielder followed up a tremendous performance in midweek with a fabulous strike as he curled an effort from the edge of the box into the far corner.
It was a great start for the Swans as the game remained wide open with both sides attacking at every opportunity, Dennis Lawrence forcing Nelson into another save.
But clear cut chances were in short supply after Butler's opener, however, although Swansea were giving away far too many free-kicks for comfort on the half-hour mark.
Paul Anderson and Butler continued to look dangerous out wide as Anderson forced Nelson to tip a sweet effort over the bar.
Then Butler created a chance for Feeney in the 40th minute with a neat cross, but Feeney's header was saved on the line by Nelson.
A mistake from Marcos Painter at the other end let in Melligan, but he decided to take his shot early and it flew wide.
Pratley was booked just before the break for unsporting behaviour as Orient forced three consecutive corners in injury time.
But Swansea's defence held strong to take a deserved one-goal lead in at half-time having dominated most of the game.
Half-time: Leyton Orient 0 Swans 1
The Swans got off to the perfect start after the break as they added a second after just 15 seconds. Bodde's shot was blocked and Darren Pratley followed up to fire a super dipping shot over Nelson from all of 30 yards.
Swansea were flying now and Tom Butler grabbed his second with a low shot with 51 minutes gone.
Six minutes later and Orient were stunned when Swansea flew into a 4-0 lead when Paul Anderson cut inside to fire another low shot beyond Nelson.
Swansea were brimming with confidence as the home fans started to drift away from the Matchroom Stadium as Nelson was forced to pull off another two saves from Bodde and Butler.
But the visitors weren't finished and two minutes into injury time, Warren Feeney made it five with his second goal in as many games to round off an unbelievable result.
